If you want to use overboard discharge at least 25 miles out do both sea cocks have to be opened?

Only the outlet needs to be open.

If you are unfamiliar with the discharge system or unfamiliar with that particular boat, be sure to check that the holding tank vent is clear before pumping out. Most of the modern boats use a polypropylene material for their tanks and if the vent is plugged by insects or kinked hose the pump may collapse the tank if the vent is plugged. Just a heads up if you haven't "been there before". Many good quality diaphragm pumps that are sometimes used on these systems will pull a considerable vacuum before stalling. Enough to damage a thin wall plastic tank.
